NF-1 Antibody (D-2) is a mouse monoclonal IgG1 kappa light chain antibody that detects NF-1 protein of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry, and enzyme-linked immunosorbent assay (ELISA). NF-1 (D-2) antibody is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. NF-1, also known as CTF, plays a crucial role in regulating gene expression by binding to specific DNA sequences, thereby stimulating DNA replication and activating transcription. NF-1 (D-2) monoclonal antibody recognizes a protein that is involved in the transcriptional regulation of numerous genes essential for cellular processes, including cell growth and differentiation. NF-1 has been shown to interact with the adenovirus type 2 promoter and activates transcription of the herpes simplex virus thymidine kinase genes. The NF-1 consensus element is found in the upstream promoter regions of various eukaryotic genes, such as Ha-Ras, α-globin, HSP 70, GRP 78, and myelin basic protein, highlighting its significance in diverse biological pathways. Two forms of NF-1, resulting from alternative splicing of a single gene, further underscore the complexity and regulatory potential of this transcription factor in cellular function.

Are there any publications showing successful ChIP experiments using the TransCruz version of this antibody?

Thank you for your question. This is a relatively new monoclonal antibody and we are not aware of any publications using this antibody for ChIP at this time.

This antibody is stated to recognize all isoforms of NF-1. Does this mean it recognize all isoforms of one of the four NF-1 genes (i.e. NF1-A, -B, -C and -X) or that it recognizes all four different NF-1 genes (NF1-A, -B, -C and -X)?

Thank you for your question. NF-1 Antibody (D-2): sc-74444 is a mouse monoclonal raised against amino acids 1-300 of NF1C of human origin. Based on sequence analysis, the antibody is expected to react with four different NF-1 genes (NFIA, NFIB, NFIC and NFIX). The protein sequences are highly conserved in this region.

When should the TransCruz reagent sc-74444 X be used instead of the standard form sc-74444?

Thank you for your question. We recommend using the TransCruz reagent form when performing ChIP or GelShift assays. This form can also be used in other applications such as Western Blot or IHC, but the dilution should be adjusted by a factor of 10 to account for the higher original concentration.
