NF-1 Antibody (A-12) is a mouse monoclonal IgG1 antibody that detects NF-1 in mouse, rat, and human samples through applications such as western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry, and enzyme-linked immunosorbent assay (ELISA). NF-1, also known as CTF, is a crucial member of the CCAAT box binding protein family, playing a significant role in stimulating DNA replication and activating transcription. NF-1 binds to specific DNA elements as a homodimer, utilizing an amino-terminal DNA binding domain, and activates transcription through a unique proline-rich carboxy-terminal transactivation domain. NF-1 has been shown to interact with the adenovirus type 2 promoter and is involved in the transcriptional activation of herpes simplex virus thymidine kinase genes. The consensus element recognized by NF-1 is present in the upstream promoter regions of numerous eukaryotic genes, including those for Ha-Ras, α-globin, HSP 70, GRP 78, Histone H1, and myelin basic protein, as well as in the promoter of the Xenopus laevis vitellogenin gene. Anti-NF-1 antibody (A-12) versatility in various applications makes this reagent an essential tool for researchers studying gene regulation and protein interactions in diverse biological contexts.

what isoform of NF1 detected by this antibody? why NF1 protein shown in WB is about 70K as it is know NF1 protein is about 300K in molecular weight.

Asked by: Anna
Thank you for your question. NF1 has a calculated molecular weight of approximately 47 kDa and is observable at higher molecular weights in western blots due to a number of post-translational modifications. The antibody is capable of detecting all NF-1 isoforms of mouse, rat and human origin by WB, IP, IF, IHC(P) and ELISA.

When should the TransCruz reagent sc-74445 X be used instead of the standard form sc-74445?

Asked by: cjMara
Thank you for your question. We recommend using the TransCruz reagent form when performing ChIP or GelShift assays. This form can also be used in other applications such as Western Blot or IHC, but the dilution should be adjusted by a factor of 10 to account for the higher original concentration.

Is this against NFI (Nuclear Factor I) or NF-1 (neurofibromin-1)?  If the former, which family member NFIA, NFIB, NFIC or NFIX?

Asked by: rich
Thank you for your question. This antibody detects NF-1, Nuclear Factor I, and it will detect all the isoforms. If you go to the product block and click on the "gene info" tab, you can see which isoforms it detects (NFIA, NFIB, NFIC and NFIX).
