Neutrophil Elastase Antibody (G-2) is a mouse monoclonal IgG1 kappa light chain antibody designed to recognize amino acids 211-267 of human Neutrophil Elastase (NE), also known as ELANE or medullasin. Anti-NE antibody (G-2) effectively detects NE through applications such as western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry with paraffin-embedded sections (IHCP), and enzyme-linked immunosorbent assay (ELISA), making this tool essential for various research settings. NE is a serine protease predominantly stored in azurophilic granules of neutrophils, where NE plays a critical role in degrading phagocytosed pathogens and remodeling the extracellular matrix during immune responses. Post-translational modifications of NE, including glycosylation, are crucial for proper folding, stability, and enzymatic activity, ensuring effective function without causing unintended tissue damage. These modifications also regulate NE′s interactions with other proteins in the complement system and role as a platelet agonist, enhancing platelet aggregation, secretion, and calcium mobilization. The structural integrity maintained by these modifications is vital for NE′s involvement in inflammatory diseases such as idiopathic pulmonary fibrosis, rheumatoid arthritis, adult respiratory distress syndrome, and cystic fibrosis. By specifically targeting modified regions of NE, NE monoclonal antibody (G-2) provides reliable and accurate detection, facilitating advanced studies into molecular mechanisms of inflammation and tissue injury. Available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates, NE (G-2) antibody accommodates a wide range of experimental protocols.

Which antigen retrival method you tried with that specific antibody? Did it work with Tris-EDTA pH 9.0?

How can Neutrophil Elastase (G-2): sc-55549 monoclonal antibody be used for double or triple staining, if the other primary antibodies are also raised in mouse?

In this case, we recommend using a directly conjugated mouse monoclonal primary antibody. This mouse monoclonal antibody, Neutrophil Elastase (G-2), is currently available conjugated to phycoerythrin (sc-55549 PE), fluorescein (sc-55549 FITC), Alexa Fluor® 488 (sc-55549 AF488) or Alexa Fluor® 647 (sc-55549 AF647)

Can you recommend a positive control to use with Neutrophil Elastase (G-2): sc-55549 monoclonal antibody for Western blot?

We recommend U-937 (sc-2239) or HL-60 (sc-2209) whole cell lysates to use as positive controls as we have Western blot data showing specific detection of Neutrophil Elastase in these cell lines.

Neutrophil Elastase expressed in the activated neutrophil in the lysosomes. Azurophil granules stained with alexa 488. Very good antibody recognise human and mouse neutrophil.
