Neurabin-II Antibody (D-7) is a mouse monoclonal IgG1 kappa light chain antibody that detects Neurabin-II protein of human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry with paraffin-embedded sections (IHCP), and enzyme-linked immunosorbent assay (ELISA). Neurabin-II (D-7) antibody is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. Neurabin-II, also known as spinophilin, plays a crucial role in the central nervous system by interacting with actin and protein phosphatase 1 (PP-1) within dendritic spines. The gene encoding human Neurabin-II is located on chromosome 17q21-q22, and structural features include an F-actin binding domain at the N-terminus, a coiled-coil structure at the C-terminus, a PDZ domain in the middle region, and a domain that interacts with transmembrane proteins. These structural characteristics enable Neurabin-II to bundle actin filaments in vitro and localize to cortical sites of actin filaments and areas of active membrane remodeling in vivo. Furthermore, Neurabin-II forms a complex with the catalytic subunit of PP-1, modulating enzymatic activity, which is essential for regulating the activity of α-amino-3-hydroxy-5-methyl-4-isoxazolepropionic acid (AMPA) and N-methyl-D-aspartate (NMDA) receptors. By doing so, Neurabin-II influences both glutamatergic synaptic transmission and dendritic morphology, highlighting Neurabin-II′s importance in synaptic plasticity and neuronal communication. Additionally, the synergistic interactions between spinophilin and the human tumor suppressor ARF suggest a potential role for Neurabin-II in cell growth regulation, making Neurabin-II a valuable target for research in neurobiology and cancer studies.

Why does neurabin II run at ~130kD, when the sequence predicts an 89kDa protein? Seems like a large difference.

Thank you for your question. The observed molecular weight is significantly higher than the predicted molecular weight. Please refer to the following publication as a reference for an approximately 140 kDa observed molecular weight for Neurabin-II, also known as Spinophilin, PMID: 9275233.

I am using Neurabin-II (D-7): sc-373974 monoclonal antibody to stain paraffin embedded tissue. Do you recommend antigen retrieval when using this antibody?

Rated 5 out of 5 by from Great IF staining cortical neuronsStains cultured cortical neurons beautifully. Looks just like actin stain in the growth cone. First SPN antibody I have had that works. These are 36hr cultures of mouse cortical neurons. (Yes, it works on mouse) Use methanol fixation- (I do mine after PFA fixation) and the antibody at 1:100. Without methanol you can see a bit but its super faint.
