NEDD4-L Antibody (C-12) is a mouse monoclonal IgG1 antibody that detects NEDD4-L in mouse, rat, and human samples through applications such as western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). NEDD4-L, also known as neural precursor cell expressed, developmentally down-regulated 4-like, RSP5, NEDD4-2, or NEDL3, is a 975 amino acid protein predominantly localized in the cytoplasm, featuring four WW domains, one HECT domain, and one C2 domain. NEDD4-L functions as an E3 ubiquitin-protein ligase, facilitating the transfer of ubiquitin from an E2 ubiquitin-conjugating enzyme to specific substrate proteins, which are then marked for proteasomal degradation. By ubiquitinating key proteins such as Smad2 and TGFβ RII, NEDD4-L inhibits the TGFβ signaling pathway, a critical regulator of cell growth and differentiation. NEDD4-L′s ability to modulate these pathways highlights its importance in various cellular processes and potential implications in diseases where TGFβ signaling is dysregulated. NEDD4-L is expressed as eight isoforms due to alternative splicing, contributing to functional diversity and regulatory mechanisms within different tissues, particularly in the pancreas, prostate, and kidney where expression is highest.
