Myocilin is an extracellular protein predominantly expressed in the eye, particularly in the retina, trabecular meshwork, and ciliary body, where myocilin plays a crucial role in maintaining intraocular pressure (IOP) homeostasis. Myocilin forms homomultimers in vivo and interacts with components of the extracellular matrix, notably through its Hep II domain, which binds to fibronectin. Additionally, myocilin associates with myosin regulatory light chain, a key component of the myosin motor protein complex, suggesting myocilin′s involvement in the actomyosin system. This interaction links myocilin to the functional status of the trabecular meshwork, which is essential for regulating IOP.
