MYLK Antibody (MLJ-9) is a mouse monoclonal IgG1 antibody that detects myosin light chain kinase (MYLK) in human samples through western blotting (WB). MYLK is a crucial enzyme that plays a significant role in muscle contraction by phosphorylating myosin regulatory light chains, which facilitates the interaction between myosin and actin filaments, ultimately leading to contractile activity. This process is vital not only in muscle tissues but also in various non-muscle cells, where MYLK isoforms contribute to cellular functions such as motility and shape maintenance. The MYLK gene encodes multiple isoforms, including those specific to smooth muscle and non-muscle tissues, as well as telokin, a unique isoform that stabilizes unphosphorylated myosin filaments in smooth muscle. The presence of different MYLK isoforms across a wide range of adult and fetal tissues underscores MYLK′s importance in diverse physiological processes. MYLK interacts with p21-activated kinases (PAKs), which can modulate MYLK activity; for instance, the interaction with PAK1 leads to a decrease in MYLK activity and subsequent myosin light chain phosphorylation, highlighting the regulatory mechanisms that control muscle contraction and cellular dynamics.
