MX1 encodes myxovirus resistance protein 1 (Mx1), a dynamin-like interferon-stimulated GTPase that mediates intrinsic antiviral defense by restricting replication of diverse RNA viruses. Mx1 is induced downstream of type I and type III interferon signaling through the JAK–STAT pathway and integrates into broader innate immune programs governed by ISGF3-driven transcription. By modulating viral nucleocapsid trafficking and replication complex integrity, Mx1 contributes to cellular stress responses that shape cytokine output and immune cell–intrinsic resistance. Dysregulated MX1 expression is widely used as a molecular readout of interferon pathway activation and is relevant to studies of viral susceptibility, chronic inflammatory states, and tumor microenvironments characterized by elevated interferon signatures.
Mx1 CRISPR/Cas9 KO Plasmid (h) is a pool of plasmids designed for targeted disruption of the MX1 gene in human cell lines. Each plasmid co-expresses a unique single guide RNA (sgRNA) targeting a distinct site within the MX1 together with the Streptococcus pyogenes Cas9 nuclease. The plasmids also encode GFP, allowing fluorescent identification and enrichment of successfully transfected cells by fluorescence microscopy or flow cytometry.
The multi-guide design increases the likelihood of generating insertions or deletions (indels) that disrupt the MX1 open reading frame following Cas9-mediated double-strand break formation. DNA breaks introduced by the CRISPR/Cas9 system are repaired through endogenous non-homologous end joining (NHEJ) pathways, frequently resulting in frameshift mutations that abolish Mx1 protein expression.
This CRISPR knockout system enables efficient generation of MX1-deficient cell models for investigation of Mx1 signaling, functional genomics studies, cancer biology research, and evaluation of therapeutic responses in human cell lines.
