Mx1/2/3 Antibody (C-1) is a mouse monoclonal IgG1 kappa light chain antibody that detects Mx1/2/3 protein of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). Mx1/2/3 (C-1) antibody is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. Mx1/2/3 proteins, particularly Mx1 and Mx2, play crucial roles in immune response to viral infections, functioning as GTPases within the Dynamin family of microtubule-associated proteins. Mx2, localized in the cytoplasm, is induced by interferon-alpha and interferon-beta, leading to accumulation and subsequent inhibition of vesicular stomatitis virus replication, providing resistance to viral infection. Mx1 is found in the nucleus and, upon similar induction, offers selective resistance against highly pathogenic H5N1 influenza virus. Mx3 serves as a rat-specific member of this protein family, contributing to overall antiviral defense mechanisms. Mx1 and Mx2 localization to different cellular compartments underscores distinct yet complementary roles in immune response, making anti-Mx1/2/3 antibody (C-1) an essential tool for studying viral resistance mechanisms in various species.

We are planning to use this clone in human muscle forzen samples, but we are unable to find any article which describes the IHC protocol (pretreatment type and time, primary time). Could you provide it?

Rated 1 out of 5 by from Negative signal observed in IF in skeletal muscle1. Frozen skeletal muscle section was fixed with acetone in 10min. 2. Wash the section in PBS buffer for 3×5minutes. 3. Place setion in 3% Hydrogen peroxide /methanol for 10 minutes 4.Wash the section in PBS buffer for 3×5minutes. 5.Place setion in 10% normal goat serum for 20min 6.Incubate section with primary anitbody. Dilution:1:200 Diluent:1%BSA, 4℃,overnight 7.Wash the section in PBS buffer for 3×5minutes. 8.Incubate section with secondary anitbody. goat anti-mouse IgM ,ALEX FLOUR 555 Dilution:1:150 Diluent:10% goat serum,room temperature 40min. 9.Wash the section in PBS buffer for 3×5minutes. 10.Moutn the section and observe.
