MRG1 Antibody (JA22) is a mouse monoclonal IgG1 kappa light chain antibody that detects MRG1 protein of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and immunohistochemistry. Anti-MRG1 antibody (JA22) is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. MRG1 protein, also known as MSG1-related gene 1, plays a crucial role as a primary response gene and shares significant sequence similarity with the carboxy-terminal region of MSG1 (melanocyte-specific gene-1). Both MRG1 and MSG1 contain two conserved domains, CR1 and CR2, with CR2 being essential for transcriptional activation, indicating that they belong to a unique family of transcription factors. MRG1 is primarily located in the nucleus, where MRG1 regulates gene expression in response to various cytokines, including interleukin-1 alpha, interleukin-9, and granulocyte-macrophage colony-stimulating factor, as well as serum growth factors, through the JAK/STAT signaling pathway. Nuclear localization of MRG1 is vital for function in transcriptional regulation, as nuclear presence allows interaction with other transcription factors and co-activators, thereby influencing cellular responses to external stimuli. Notably, overexpression of MRG1 has been linked to oncogenic properties, such as anchorage-independent growth and tumor formation in nude mice, highlighting potential role in cancer biology. Additionally, a splice variant of MRG1, known as p35srj, is ubiquitously expressed and has been shown to interact with the p300-CH1 domain of p300/CBP, inhibiting interaction with hypoxia-inducible factor-1 alpha to prevent HIF-1 transactivation, further underscoring significance in cellular signaling pathways.

When should the TransCruz reagent sc-21795 X be used instead of the standard form sc-21795?

Thank you for your question. We recommend using the TransCruz reagent form when performing ChIP or GelShift assays. This form can also be used in other applications such as Western Blot or IHC, but the dilution should be adjusted by a factor of 10 to account for the higher original concentration.
