MMP-2 Antibody (4D3) is a mouse monoclonal IgG1 kappa light chain antibody, developed to recognize and bind to the amino acid residues 557-569 of the human Matrix metalloproteinase-2 (MMP-2) protein, while also demonstrating reactivity with MMP-2 from mouse, rat, and additional species including rabbit, and potential cross-reactivity with MMP-3. MMP-2 monoclonal antibody (4D3) is recommended for use in various applications such as western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and immunohistochemistry with paraffin-embedded sections (IHCP). MMP-2, also called gelatinase A, CLG4, MONA, CLG4A, and TBE-1, is a crucial enzyme within the matrix metalloproteinase (MMP) family, characterized by its complex structure comprising a signal peptide, a propeptide domain that maintains the enzyme in an inactive state, a catalytic domain containing a zinc-binding motif essential for its proteolytic activity, and a hemopexin-like domain that confers substrate specificity and mediates interactions with tissue inhibitors of metalloproteinases (TIMPs). The structural integrity of MMP-2 is vital for its function in degrading extracellular matrix (ECM) components such as collagen types IV, V, VII, and X, as well as gelatin type I, facilitating processes like tissue remodeling, angiogenesis, and wound healing. The propeptide domain must be cleaved to activate MMP-2, a process regulated by the Ras signaling pathway and dependent on the presence of calcium and zinc ions for catalytic activity. The hemopexin-like domain not only dictates substrate specificity but also plays a pivotal role in the enzyme′s interaction with inhibitors, thereby regulating activity within the cellular environment. Anti-MMP-2 antibody (clone 4D3) is recommended for researchers studying the role of MMP-2 in extracellular matrix remodeling, angiogenesis, and wound healing, as well as regulation and interactions within the matrix metalloproteinase family and inhibitors. Available in both non-conjugated and multiple conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and various Alexa Fluor® conjugates, MMP-2 (4D3) antibody caters to diverse experimental requirements.
