MLH1 Antibody (A-8) is a mouse monoclonal IgG2a antibody that detects MLH1 in mouse, rat, and human samples through applications such as western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). MLH1 plays a crucial role in the DNA mismatch repair (MMR) pathway, which is vital for maintaining genetic stability by correcting DNA replication errors. MLH1 (A-8) antibody enables detection of deficiencies that can lead to microsatellite instability and an increased risk of various cancers, including hereditary nonpolyposis colorectal cancer (HNPCC). MLH1 is predominantly located in the nucleus, where MLH1 forms a complex with other MMR proteins to identify and repair mismatched DNA bases. This localization is significant because MLH1 allows for immediate response to DNA damage, thereby preventing the accumulation of mutations that could lead to tumorigenesis. Expression of MLH1 is often lost in several cancer types, including certain ovarian tumors, which can be attributed to gene methylation or mutations. When should the TransCruz reagent sc-133228 X be used instead of the standard form sc-133228?

Thank you for your question. We recommend using the TransCruz reagent form when performing ChIP or GelShift assays. This form can also be used in other applications such as Western Blot or IHC, but the dilution should be adjusted by a factor of 10 to account for the higher original concentration.
