MLH1 Antibody (A-8) is a mouse monoclonal IgG2a antibody that detects MLH1 in mouse, rat, and human samples through applications such as western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). MLH1 plays a crucial role in the DNA mismatch repair (MMR) pathway, which is vital for maintaining genetic stability by correcting DNA replication errors. MLH1 (A-8) antibody enables detection of deficiencies that can lead to microsatellite instability and an increased risk of various cancers, including hereditary nonpolyposis colorectal cancer (HNPCC). MLH1 is predominantly located in the nucleus, where MLH1 forms a complex with other MMR proteins to identify and repair mismatched DNA bases. This localization is significant because MLH1 allows for immediate response to DNA damage, thereby preventing the accumulation of mutations that could lead to tumorigenesis. Expression of MLH1 is often lost in several cancer types, including certain ovarian tumors, which can be attributed to gene methylation or mutations. Anti-MLH1 antibody (A-8) is particularly useful for researchers studying the implications of MMR deficiencies in cancer biology and therapeutic responses, as MLH1 (A-8) monoclonal antibody provides a reliable means to assess MLH1 expression levels in various tissues and cell lines.
