mHMGCS Antibody (B-8) is a mouse monoclonal IgG2a kappa light chain antibody that detects mHMGCS protein of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry, and enzyme-linked immunosorbent assay (ELISA). anti-mHMGCS antibody (B-8) is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. mHMGCS, or mitochondrial HMG-CoA synthase, plays a crucial role in ketone body biosynthesis by catalyzing the condensation of acetyl-CoA with acetoacetyl-CoA to form HMG-CoA, which is subsequently converted into mevalonate by HMG-CoA reductase, initiating the isoprenoid biosynthesis pathway. This pathway is vital for producing essential biomolecules, including cholesterol and ubiquinone, which are critical for cellular membrane integrity and energy production, respectively. mHMGCS is predominantly expressed in liver and kidney, and expression is regulated by various metabolic states; fasting, cyclic adenosine monophosphate, and fatty acids enhance transcription, while feeding and insulin exert a repressive effect. Additionally, a regulatory element within mHMGCS promoter allows for transcriptional regulation by nuclear receptors such as PPAR, RXR, COUP-TF, and HNF-4, highlighting mHMGCS′s importance in metabolic regulation and energy homeostasis.
