MGMT Antibody (MT 3.1) is a mouse monoclonal IgG1 kappa light chain antibody raised against MGMT of human origin, showing reactivity with MGMT from human and, to a lesser extent, mouse and rat samples by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry with paraffin-embedded sections (IHCP), and flow cytometry (FCM). MGMT, also known as O6-methylguanine-DNA methyltransferase or O-6-alkylguanine DNA alkyltransferase (AGT), is a crucial DNA repair enzyme predominantly located in the cell nucleus. Nuclear localization of MGMT allows direct access to damaged DNA, where anti-MGMT antibody (MT 3.1) removes mutagenic and cytotoxic O6-alkylguanine lesions caused by carcinogens or cytostatic drugs, thus maintaining genomic stability. MGMT activity is essential for protecting cells from DNA alkylation damage, and MGMT expression levels can influence the effectiveness of certain chemotherapeutic agents. High MGMT expression is often associated with resistance to alkylating drugs. MGMT expression is transcriptionally activated in response to DNA damage, and this induction requires functional p53, a tumor suppressor protein involved in DNA repair and apoptosis. Studies have shown that high MGMT activity correlates with mutant p53 in some cancer cell lines, suggesting a complex regulatory relationship. Understanding cellular localization and regulation of MGMT is important for cancer research and therapeutic development, making MGMT monoclonal antibody (MT 3.1) a valuable tool for researchers investigating DNA repair mechanisms and p53-related pathways.
