MetAP-1, a member of the M24 family of proteins, plays a crucial role in protein maturation by removing the amino-terminal methionine residue from nascent polypeptides, a process essential for proper protein folding and function. This 394-amino acid protein is expressed at low levels across various tissues but is particularly abundant in skeletal muscle, highlighting MetAP-1′s importance in muscle physiology. The active site of MetAP-1 is characterized by the presence of two adjacent divalent metal ions, which are crucial for enzymatic activity, as these ions are connected by a water molecule or hydroxide ion.
