Megalin Antibody (H-10) is a mouse monoclonal IgG1 kappa light chain antibody that detects megalin in mouse, rat, and human samples through various applications including western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry with paraffin-embedded sections (IHCP), and enzyme-linked immunosorbent assay (ELISA). Megalin (H-10) antibody is available in both non-conjugated and multiple conjugated forms, such as agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and various Alexa Fluor® conjugates. Megalin, also known as GP330, is a member of the low-density lipoprotein receptor gene family, which includes other receptors like LDLR, LRP, VLDLR, and ApoER2. Megalin is primarily located on the apical membrane of epithelial cells, particularly in the proximal kidney tubules, intestine, and ependymal cells, where megalin plays a crucial role in the reabsorption of essential molecules, including vitamin B12. Proper localization of megalin is vital for mediating the uptake of various ligands, such as apolipoprotein J (also known as Clusterin), which is associated with the clearance of beta-amyloid peptides linked to Alzheimer′s disease. Additionally, megalin serves as an antigenic determinant in Heymann nephritis in rats, highlighting megalin′s significance in kidney function and disease.
