MCT4 Antibody (D-1) is a mouse monoclonal IgG2a kappa light chain antibody engineered to detect Monocarboxylate Transporter 4 (MCT4) in mouse, rat, and human species through applications such as western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry with paraffin-embedded sections (IHCP), and enzyme-linked immunosorbent assay (ELISA). Anti-MCT4 antibody (D-1) specifically targets amino acids 376-465 within the C-terminal cytoplasmic domain of human MCT4, ensuring precise binding and reliable detection. MCT4 is a member of the solute carrier 16A (SLC16A) family of proteins, and is an integral membrane protein characterized by 10-12 transmembrane-helical domains, with both amino and carboxy termini located in the cytoplasm. This structural configuration facilitates proton-linked transport of lactate and other monocarboxylates across the plasma membrane. MCT4 (D-1) antibody plays a vital role in studying intracellular pH balance and sustained glycolytic activity, particularly in highly glycolytic tissues such as skeletal muscle and cancer cells where rapid lactate production is driven by the Warburg effect. Strategic localization of MCT4 on the plasma membrane effectively manages lactate levels, preventing intracellular acidification and supporting cellular metabolism and survival under anaerobic conditions. Understanding MCT4 structure underpins research into cancer metabolism and other diseases characterized by altered lactate transport. Anti-MCT4 antibody (D-1) is available in non-conjugated form and various conjugates, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates, providing versatile options for diverse experimental needs. MCT4 monoclonal antibody (D-1) supports research on expression, localization, and functional roles of MCT4 in various physiological and pathological contexts.

I am planning to use this antibody for detecting mouse MCT4 by FCM. Can you confirm that this antibody specifically targets the cytoplasmic domain of MCT4 and is suitable for use in both mouse and human samples?

Which specific antigen retrieval do you recommand using MCT4 (D-1): sc-376140 antibody between :10 mM sodium citrate buffer, pH 6.0 (sc-294091); 50 mM glycine-HCl buffer (sc-29096), pH 3.5, and 0.01% (w/v) EDTA (sc-29092)?

Thank you for your question. For antigen retrieval in experiments with this antibody we recommend heat treatment: Heat treatment (recommended method): Place slides in a container and cover with 10 mM sodium citrate buffer, pH 6.0 (sc-294091); or with 50 mM glycine-HCl buffer (sc-29096), pH 3.5, with 0.01% (w/v) EDTA (sc-29092). Heat at 95° C for 5 minutes. Top off with fresh buffer and heat at 95° C for 5 minutes (optimal incubation time may vary for each tissue type). Allow slides to cool in the buffer for approximately 20 minutes. Wash in deionized H2O three times for 2 minutes each. Aspirate excess liquid from slides.

I tried on mouse sample (4T1 cells lysate ) for WB, but doesn't work. is it possible to have the an aliquot of the discontinued MCT4 (sc-50329) that was working in my previous experiments?
