
Mcl-1 Antibody (RC13) is a mouse monoclonal IgG1 kappa light chain antibody, raised against amino acids 1-327 of human Mcl-1 protein, and is recommended for detection of Mcl-1 of human and, to a lesser extent, mouse and rat origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and immunohistochemistry with paraffin-embedded sections (IHCP). Mcl-1, also called Myeloid cell leukemia-1, Mcl1, and Gm52627, is a critical member of the Bcl-2 family of proteins that regulate apoptosis, the programmed cell death essential for development and tissue homeostasis. Predominantly localized to mitochondria, Mcl-1 plays a vital role in inhibiting apoptosis by binding to pro-apoptotic proteins like Bak and Bax, preventing mitochondrial outer membrane permeabilization and cytochrome c release, which would otherwise trigger cell death pathways. Mcl-1′s mitochondrial localization is key to function, as Mcl-1 directly influences cell survival by regulating mitochondrial integrity. Dysregulation or overexpression of Mcl-1 is linked to various cancers, including leukemia and lymphoma, due to Mcl-1′s role in allowing malignant cells to evade apoptosis. Mcl-1 stability is modulated by glycogen synthase kinase 3 (GSK3), which affects apoptosis regulation through pathways like PI 3-kinase and Akt. Studies in Mcl-1-deficient mice show significant reductions in B and T lymphocytes, underlining Mcl-1′s importance in early hematopoietic progenitors.
