
Mcl-1 Antibody (MCL701) is a mouse monoclonal IgG1 kappa light chain antibody specifically designed to detect Mcl-1 proteins from human origin. anti-Mcl-1 antibody (MCL701) targets the full-length recombinant Mcl-1, enabling reliable detection in human samples through various applications such as western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry with paraffin-embedded sections (IHCP), and enzyme-linked immunosorbent assay (ELISA). Mcl-1, also called Myeloid cell leukemia-1, Mcl1, and Gm52627, is a pivotal anti-apoptotic member of the Bcl-2 family that resides primarily in the mitochondria, where Mcl-1 plays a crucial role in regulating cell survival and apoptosis. By inhibiting pro-apoptotic proteins like Bak and Bax, Mcl-1 prevents mitochondrial outer membrane permeabilization and the subsequent release of cytochrome c, thereby safeguarding cells from programmed cell death. The mitochondrial localization of Mcl-1 is essential for function, as this allows immediate interaction with key apoptotic regulators at the site where apoptotic signals are initiated. Additionally, Mcl-1 undergoes tight regulation through various post-translational modifications, including phosphorylation by Glycogen Synthase Kinase 3 (GSK3), which targets Mcl-1 for ubiquitination and proteasomal degradation, a process modulated by survival signals from the PI3K/Akt pathway. Mcl-1 also interacts with BH3-only proteins such as Bim, Noxa, and Puma, which can neutralize anti-apoptotic activity, and with other Bcl-2 family members like Bcl-xL and Bcl-w, maintaining a delicate balance between cell survival and apoptosis. In cancer research, overexpression of Mcl-1 is often associated with resistance to chemotherapy and poor prognosis in various malignancies, including leukemia, lymphoma, and solid tumors. Furthermore, studies in mice have demonstrated that Mcl-1 is essential for the survival of early hematopoietic progenitors, as Mcl-1 deficiency leads to early embryonic lethality and significant reductions in B and T lymphocytes, highlighting an indispensable role in cellular homeostasis and immune function.
