Mcl-1 Antibody (G-7) is a mouse monoclonal IgG2a kappa light chain antibody raised against amino acids 1-260 of human Mcl-1, suitable for detecting Mcl-1 proteins from mouse, rat, and human origins via western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry with paraffin-embedded sections (IHCP), and enzyme-linked immunosorbent assay (ELISA) applications. Mcl-1 (Myeloid cell leukemia 1) is a critical anti-apoptotic protein within the B-cell lymphoma 2 (Bcl-2) family, playing a vital role in cell survival by inhibiting apoptosis through interactions with several key proteins. Mcl-1 monoclonal antibody (G-7) binds to pro-apoptotic members such as Bak (Bcl-2 antagonist killer) and Bax (Bcl-2-associated X protein), sequestering them and preventing mitochondrial outer membrane permeabilization and cytochrome c release, which are essential steps in the intrinsic apoptotic pathway. Anti-Mcl-1 antibody (G-7) also interacts with BH3-only proteins like Bim (Bcl-2-interacting mediator of cell death), NOXA, and PUMA, which can bind to Mcl-1 and neutralize anti-apoptotic function, thereby promoting apoptosis when necessary. Stability and expression of Mcl-1 are regulated by interactions with E3 ubiquitin ligases such as Mcl-1 ubiquitin ligase E3 (MULE) and β-TrCP, targeting for ubiquitination and proteasomal degradation. Glycogen synthase kinase 3 (GSK3) phosphorylates Mcl-1, facilitating recognition by these ubiquitin ligases. Additionally, tumor suppressor p53 can influence Mcl-1 levels by promoting expression of pro-apoptotic proteins that antagonize Mcl-1 function. This intricate network of interactions involving Mcl-1 is crucial for maintaining cellular homeostasis, and dysregulation can lead to diseases such as cancer, where overexpression of Mcl-1 contributes to tumor progression and resistance to chemotherapy. Mcl-1 (G-7) antibody serves as an invaluable tool for researchers investigating molecular mechanisms of apoptosis, protein-protein interactions involving Mcl-1, and implications in human diseases.

I have used this antibody at 1:1000 dilution. But it didn't work and some references have used this antibody at 1:100 in WB. Which conc. should I try?

Thank you for your question. We recommend using this antibody, Mcl-1 Antibody (G-7): sc-74437, at a starting dilution of 1:200 for Western blot. The recommended range is 1:100-1:1,000 for testing to determine the optimal concentration to be used on your samples.

I am using Mcl-1 (G-7): sc-74437 monoclonal antibody to stain paraffin embedded tissue. Do you recommend antigen retrieval when using this antibody?

Rated 1 out of 5 by from Not good for my IFI performed IF using free sample of this antibody in my RPE1 cells. Unfortunately, all 3 ways of my fixation&permeabilisation (4% PFA fixing+0.5% Triton permeabilisation, 4% PFA fixing+MeOH permeabilisation and MeOH fixing) didn't work well at all with this antibody. This Mcl-1 antibody seems good for western though not the best for IF.
