
Mcl-1 Antibody (G-7) is a mouse monoclonal IgG2a kappa light chain antibody raised against amino acids 1-260 of human Mcl-1, suitable for detecting Mcl-1 proteins from mouse, rat, and human origins via western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry with paraffin-embedded sections (IHCP), and enzyme-linked immunosorbent assay (ELISA) applications. Mcl-1 (Myeloid cell leukemia 1) is a critical anti-apoptotic protein within the B-cell lymphoma 2 (Bcl-2) family, playing a vital role in cell survival by inhibiting apoptosis through interactions with several key proteins. Mcl-1 monoclonal antibody (G-7) binds to pro-apoptotic members such as Bak (Bcl-2 antagonist killer) and Bax (Bcl-2-associated X protein), sequestering them and preventing mitochondrial outer membrane permeabilization and cytochrome c release, which are essential steps in the intrinsic apoptotic pathway. Anti-Mcl-1 antibody (G-7) also interacts with BH3-only proteins like Bim (Bcl-2-interacting mediator of cell death), NOXA, and PUMA, which can bind to Mcl-1 and neutralize anti-apoptotic function, thereby promoting apoptosis when necessary. Glycogen synthase kinase 3 (GSK3) phosphorylates Mcl-1, facilitating recognition by these ubiquitin ligases. Additionally, tumor suppressor p53 can influence Mcl-1 levels by promoting expression of pro-apoptotic proteins that antagonize Mcl-1 function.
