Neurod6 encodes the basic helix–loop–helix transcription factor MATH-2, a key regulator of neuronal differentiation, maturation, and survival in the developing mouse nervous system. MATH-2 influences gene programs that support neurite outgrowth, axon guidance, and maintenance of glutamatergic neuronal identity, coordinating with other proneural bHLH factors to shape cortical and hippocampal circuitry. Through transcriptional control of neuronal fate and connectivity, Neurod6 is relevant to studies of neurodevelopmental mechanisms and pathways implicated in cognitive and behavioral phenotypes. Dysregulated bHLH-driven differentiation networks are commonly examined in models of neurodevelopmental disorders and neuronal vulnerability.
MATH-2 CRISPR/Cas9 KO Plasmid (m) is a pool of plasmids designed for targeted disruption of the Neurod6 gene in mouse cell lines. Each plasmid co-expresses a unique single guide RNA (sgRNA) targeting a distinct site within the Neurod6 together with the Streptococcus pyogenes Cas9 nuclease. The plasmids also encode GFP, allowing fluorescent identification and enrichment of successfully transfected cells by fluorescence microscopy or flow cytometry.
The multi-guide design increases the likelihood of generating insertions or deletions (indels) that disrupt the Neurod6 open reading frame following Cas9-mediated double-strand break formation. DNA breaks introduced by the CRISPR/Cas9 system are repaired through endogenous non-homologous end joining (NHEJ) pathways, frequently resulting in frameshift mutations that abolish MATH-2 protein expression.
This CRISPR knockout system enables efficient generation of Neurod6-deficient cell models for investigation of MATH-2 signaling, functional genomics studies, cancer biology research, and evaluation of therapeutic responses in human cell lines.
