MAN1B1 Antibody (E-10) is a mouse monoclonal IgG1 kappa light chain antibody that detects MAN1B1 protein of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). MAN1B1 (E-10) antibody is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. MAN1B1 protein, also known as mannosidase alpha, class 1B, member 1, or MANA-ER, is a crucial enzyme belonging to the glycosyl hydrolase 47 family, primarily localized in the endoplasmic reticulum (ER). This single-pass type II membrane protein plays a vital role in the maturation of Asn-linked oligosaccharides on glycoproteins by catalyzing the initial mannose trimming step, which is essential for proper protein folding and function. MAN1B1′s significance extends to involvement in the endoplasmic reticulum-associated protein degradation (ERAD) pathway, targeting misfolded or unassembled glycoproteins for degradation via the ubiquitin-proteasome system, thereby maintaining cellular homeostasis. Additionally, MAN1B1 activity is calcium-dependent and can be inhibited by specific class I alpha-mannosidase inhibitors such as 1-deoxymannojirimycin and kifunensine, highlighting regulatory mechanisms in glycoprotein processing.
