MACF1-4 Antibody (D-10) is a mouse monoclonal IgG1 kappa light chain antibody that detects MACF1-4 of human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). MACF1-4 (D-10) antibody is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. MACF1-4, also known as Microtubule-Actin cross-linking factor 1, isoform 4, is a large cytoplasmic protein consisting of 5,327 amino acids, encoded by the human MACF1 gene. MACF1-4 is a member of the plakin or cytolinker family and features a complex structure that includes one actin-binding domain, two calponin-homology domains, two EF-hand domains, nineteen plectin repeats, and thirty-seven spectrin repeats. The presence of spectrin repeats is particularly significant as these contribute to MACF1-4′s ability to form a three-helix bundle, which is crucial for maintaining cytoskeletal integrity and facilitating interactions with other cytoskeletal components. MACF1-4 plays a vital role in cross-linking cytoskeletal proteins, binding intermediate filaments through N-terminal plectin repeats and microtubules at C-terminus, thereby influencing cellular architecture and stability. MACF1-4 is predominantly expressed in the heart, lung, pituitary gland, and placenta, but is notably absent in the brain, kidney, liver, pancreas, and skeletal muscle, highlighting specialized functions in specific tissues.
