LEF-1 Antibody (B-6) is a mouse monoclonal IgG1 kappa light chain antibody that detects LEF-1 in mouse, rat, and human samples through various applications including western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry, and enzyme-linked immunosorbent assay (ELISA). Anti-LEF-1 antibody (B-6) is available in both non-conjugated and multiple conjugated forms, such as agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and various Alexa Fluor® conjugates. LEF-1, a member of the TCF (T cell factor) family of transcription factors, plays a crucial role in regulating gene expression during development, particularly in lymphoid and neuronal cells. LEF-1 is characterized by a high mobility group (HMG) box, which enables DNA-binding capability. This structural feature allows LEF-1 to interact with β-catenin in the nucleus, forming complexes that activate Wnt target genes, which are vital for various cellular processes including cell proliferation and differentiation. Dysregulation of this pathway, often due to mutations in the adenomatous polyposis coli (APC) gene, can lead to β-catenin accumulation and aberrant activation of LEF-1 target genes, contributing to oncogenesis, particularly in colorectal cancer. LEF-1 (B-6) antibody′s availability in multiple formats enhances experimental versatility, making this tool essential for researchers studying Wnt signaling and its implications in development and disease.

Could this antibody work for FA fixed chromatin immunoprecipitation?

Thank you for your question. For use in ChIP we recommend using LEF-1 Anticorpo (B-6) X, as the gelatin in our standard solution could interfere with the experiment.

this antibody recognizes all LEF1 isoforms ? if it does, do you have any antibody against lef1 short isoforms (dominant negative isoforms) ? thanks !

Thank you for your question. LEF-1 Antibody (B-6): sc-374522 has been raised against amino acids 61-130 mapping near the N-terminus of LEF-1 of human origin. It recognizes isoforms 1 (canonical), 2, 5, and 6. Since the amino acid sequences 1-115 are missing in isoforms 3 and 4, and 1-70 is missing in isoform 7, the antibody may not recognize these variants. LEF-1 Antibody (B-10): sc-374412 has been raised against amino acids 61-130 mapping near the N-terminus of LEF-1 of human origin as well. Please contact our technical service for further information or to arrange for a sample of the antibodies.

I am using LEF-1 (B-6): sc-374522 monoclonal antibody to stain paraffin embedded tissue. Do you recommend antigen retrieval when using this antibody?
