Lactate dehydrogenase A (LDH-A) catalyzes the interconversion of pyruvate and lactate with concomitant NADH/NAD⁺ cycling, a key reaction supporting anaerobic glycolysis and redox homeostasis. In rat cells, LDH-A helps sustain glycolytic flux under hypoxia and high energetic demand, influencing lactate production, cytosolic pH balance, and metabolic crosstalk with the TCA cycle. LDH-A activity is tightly linked to metabolic reprogramming programs that accompany proliferation and stress adaptation, including HIF-1–regulated responses and altered NAD⁺ availability. Dysregulated LDH-A–dependent lactate metabolism has been associated with pathological contexts such as ischemic injury, inflammation, and tumor-like metabolic phenotypes, making it a useful target for mechanistic studies of metabolic signaling.
LDH-A CRISPR/Cas9 KO Plasmid (r) is a pool of plasmids designed for targeted disruption of the gene in rat cell lines. Each plasmid co-expresses a unique single guide RNA (sgRNA) targeting a distinct site within the together with the Streptococcus pyogenes Cas9 nuclease. The plasmids also encode GFP, allowing fluorescent identification and enrichment of successfully transfected cells by fluorescence microscopy or flow cytometry.
The multi-guide design increases the likelihood of generating insertions or deletions (indels) that disrupt the open reading frame following Cas9-mediated double-strand break formation.
