KV4.2 Antibody (H-5) is a mouse monoclonal IgG1 kappa light chain antibody that detects KV4.2 protein of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). Anti-KV4.2 antibody (H-5) is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. KV4.2 plays a crucial role in regulating neuronal excitability and action potential repolarization, which is essential for proper neuronal signaling and muscle contraction. Located in the plasma membrane, KV4.2 is part of the voltage-gated potassium channel family, which controls potassium ion flow across cell membranes. This ion flow maintains resting membrane potential and facilitates rapid repolarization of action potentials, influencing the frequency and timing of neuronal firing. KV4.2 forms functional channels as tetramers, composed of four pore-forming alpha subunits, and KV4.2 activity can be modulated by auxiliary beta subunits. Understanding KV4.2 dynamics and interactions with other proteins is important for elucidating mechanisms underlying various neurological disorders and developing targeted therapies.

Is there any issue in using sc-390571 for detection of the short isoform of Kv4.3 (Uniprot: Q9UK17-2)? Do you have any data showing detection of the above mentioned isoform in Western blot experiment?

Thank you for your question. We have not tested this mouse monoclonal, KV4.2/4.3 Antibody (H-5): sc-390571, to evaluate potential reactivity with the long and short isoforms of the protein. All Western blot data for this antibody is shown on our website. This antibody is raised against amino acids 406-630 of KV4.2 of human origin. Based on sequence analysis, the immunogen does share only 59% sequence identity with the short isoform of Kv4.3 (Q9UK17-2). The exact epitope recognized by this antibody has not been determined, but based on this alignment there is a possibility that the antibody could recognize both the long and short isoforms.

Are there any issues with using sc-390571: KV4.2/4.3 (H-5) mouse monoclonal primary antibody on mouse samples?

To eliminate any potential cross-reactivity of an anti-mouse secondary detection reagent,we recommend using a directly conjugated primary antibody. Rated 5 out of 5 by from Very reliable antibody for a tricky protein!The KCND2/3 (Kv4.2/4.3) gene translates into proteins of theoretical molecular weight approximately 70 and 73kDa in Rat respectively. I have tried this antibody in rat cortex and hippo-campus tissue and found a 75 kDa band which is equivalent to Kv4.3/4.2 proteins (see my blot picture). The 50 kDa band may be coming from he secondary antibody because I see that in other antibodies too occasionally (time to change the secondary I guess). the higher molecular weight bands (>250kDa) which appeared as smear and possibly post translation modified form of Kv4.2/4.3, membrane proteins are highly post transnational modified ( Glycosylated etc) which may cause a huge shift in mol weight. Another thing is that the membrane proteins need special lysis buffer, they are meant to be associated with lipids, so regular RIPA buffer may not be always good to get them in dissolved state and sometimes heating tend to precipitate membrane proteins. so someone may miss the high molecular (>250) smeared bands because of his/her choice of buffer, but in this case I have used a special detergent and did not boiled the sample. so this blot opens up new questions and have the potential for a summer student project. anyway buy this antibody and it will not disappoint you. who knows, an extra band may lead to a great discovery.
