KSR1 encodes kinase suppressor of Ras 1 (Ksr-1), a scaffold protein that organizes components of the RAF–MEK–ERK/MAPK cascade to modulate signal amplitude, duration, and subcellular localization. By coordinating MAPK module assembly downstream of receptor tyrosine kinases and RAS, Ksr-1 contributes to regulation of proliferation, differentiation, and survival decisions. KSR1 activity and expression have been linked to altered MAPK pathway output in oncogenic signaling contexts, and it has also been implicated in immune and inflammatory signaling networks that intersect with ERK-dependent transcriptional programs. These features make KSR1 a useful node for dissecting pathway wiring, feedback control, and context-specific signal propagation in human cells.
Ksr-1 CRISPR/Cas9 KO Plasmid (h) is a pool of plasmids designed for targeted disruption of the KSR1 gene in human cell lines. Each plasmid co-expresses a unique single guide RNA (sgRNA) targeting a distinct site within the KSR1 together with the Streptococcus pyogenes Cas9 nuclease. The plasmids also encode GFP, allowing fluorescent identification and enrichment of successfully transfected cells by fluorescence microscopy or flow cytometry.
The multi-guide design increases the likelihood of generating insertions or deletions (indels) that disrupt the KSR1 open reading frame following Cas9-mediated double-strand break formation. DNA breaks introduced by the CRISPR/Cas9 system are repaired through endogenous non-homologous end joining (NHEJ) pathways, frequently resulting in frameshift mutations that abolish Ksr-1 protein expression.
This CRISPR knockout system enables efficient generation of KSR1-deficient cell models for investigation of Ksr-1 signaling, functional genomics studies, cancer biology research, and evaluation of therapeutic responses in human cell lines.
