KLF6 encodes Krüppel-like factor 6, a zinc-finger transcription factor that binds GC-rich promoter elements to regulate gene programs controlling cell-cycle progression, differentiation, and tissue remodeling. It integrates signals from pathways such as TGF-β/SMAD and broader stress-responsive transcriptional networks to modulate genes involved in proliferation and extracellular matrix regulation. Altered KLF6 expression or function has been associated with dysregulated growth control and inflammatory or fibrotic processes, making it relevant for studying transcriptional rewiring in disease contexts. In human cell models, KLF6 serves as a node for probing how transcription factors coordinate phenotype changes through promoter occupancy and downstream gene expression.
KLF6 CRISPR/Cas9 KO Plasmid (h) is a pool of plasmids designed for targeted disruption of the KLF6 gene in human cell lines. Each plasmid co-expresses a unique single guide RNA (sgRNA) targeting a distinct site within the KLF6 together with the Streptococcus pyogenes Cas9 nuclease. The plasmids also encode GFP, allowing fluorescent identification and enrichment of successfully transfected cells by fluorescence microscopy or flow cytometry.
The multi-guide design increases the likelihood of generating insertions or deletions (indels) that disrupt the KLF6 open reading frame following Cas9-mediated double-strand break formation. DNA breaks introduced by the CRISPR/Cas9 system are repaired through endogenous non-homologous end joining (NHEJ) pathways, frequently resulting in frameshift mutations that abolish KLF6 protein expression.
This CRISPR knockout system enables efficient generation of KLF6-deficient cell models for investigation of KLF6 signaling, functional genomics studies, cancer biology research, and evaluation of therapeutic responses in human cell lines.
