K-Ras Antibody (F234) is a mouse monoclonal IgG2a kappa light chain antibody, specifically raised against recombinant K-Ras protein encompassing amino acids 54-189 of rat origin, and demonstrates reactivity with K-Ras proteins from mouse, rat, and human sources in applications such as western blotting (WB), immunoprecipitation (IP), and immunofluorescence (IF). The K-Ras protein is a small GTPase belonging to the Ras family, playing a pivotal role in cellular signal transduction pathways that regulate cell proliferation, differentiation, and survival. K-Ras cycles between an active GTP-bound state and an inactive GDP-bound state, a process regulated by guanine nucleotide exchange factors (GEFs) and GTPase-activating proteins (GAPs). K-Ras is primarily localized to the inner surface of the plasma membrane, a location critical for function because K-Ras facilitates interactions with upstream activators and downstream effectors in signaling pathways. This membrane association is mediated by post-translational modifications, such as farnesylation and palmitoylation, which anchor K-Ras to the lipid bilayer. The precise cellular localization of K-Ras is essential for transmitting signals that control vital cellular processes, as mislocalization can disrupt normal signaling, leading to uncontrolled cell growth and contributing to oncogenesis. Mutations in the KRAS gene are among the most common in human cancers and are associated with poor prognosis. Understanding the localization and function of K-Ras is therefore crucial for cancer research and the development of targeted therapies. K-Ras (F234) antibody is an invaluable tool for researchers studying the expression, localization, and function of K-Ras in both normal physiology and disease states.

Can this SC-30 antibody be used for flow cytometry purposes? And if so, what protocol would you recommend using (in terms of obtaining the appropriate amount of antibody for flow sorting human cancer cells)?

Can anti-kras antibodies be used during immunofluorescence assays of cell cultures without a permeabilization step? In other words, can k-ras be detected and recognized from outside the cell?

Thank you for your question. K-Ras is generally a cytoplasmic membrane protein, which requires the antibody to be able to enter the cell. We recommend methanol fixation as this partially permeabilizes the cell allowing for good detection. IS THIS CAN BE USE FOR IHC AT HUMAN TISSUE SAMPLE (PARAFINATED)

Thank you for your question. We have not tested this mouse monoclonal, K-Ras (F234): sc-30, for IHC on formalin-fixed paraffin-embedded tissue sections. However, this particular clone has been cited in several publications for this research application so it may be suitable. As a reference, please refer to the following product citations on PubMed, PMID's: 12727811, 10884918, 18973197 and 19545448.

Can I use this mouse monoclonal antibody for immunohistochemical staining of mouse tissue?

No, since this antibody doesn't have any direct conjugates available, it would not be recommended for IHC in mouse tissue. See pan Ras (C-4): sc-166691 for K-Ras antibody conjugates, including AC, HRP, FITC, PE, Alexa Fluor® 488 and Alexa Fluor® 647.

hi, what dilution of this antibody to be used for western blot?

Rated 3 out of 5 by from KRas WBWe bought this antibody to do Western blots for endogenous KRas in HEK and HeLa cells. Endogenous KRas levels in those cell lines are rather low so while we can detect it, there are several strong non-specific protein bands, which is slightly dispappointing. Otherwise, not a bad antibody.
