MICALL2 encodes JRAB (also known as MICAL-L2), a Rab13 effector that functions as a scaffold linking vesicle trafficking machinery to actin cytoskeleton remodeling at cell–cell junctions. JRAB participates in regulation of adherens and tight junction organization, membrane recycling, and polarized transport, influencing epithelial barrier integrity and directional migration. Through interactions with Rab GTPases and junctional components, MICALL2 contributes to pathways governing cytoskeletal dynamics and intracellular transport. Altered expression or regulation of MICALL2 has been reported in studies of tumor cell invasion and metastasis-related phenotypes, making it relevant for investigating mechanisms of cell polarity loss and motility.
JRAB CRISPR/Cas9 KO Plasmid (h) is a pool of plasmids designed for targeted disruption of the MICALL2 gene in human cell lines. Each plasmid co-expresses a unique single guide RNA (sgRNA) targeting a distinct site within the MICALL2 together with the Streptococcus pyogenes Cas9 nuclease. The plasmids also encode GFP, allowing fluorescent identification and enrichment of successfully transfected cells by fluorescence microscopy or flow cytometry.
The multi-guide design increases the likelihood of generating insertions or deletions (indels) that disrupt the MICALL2 open reading frame following Cas9-mediated double-strand break formation. DNA breaks introduced by the CRISPR/Cas9 system are repaired through endogenous non-homologous end joining (NHEJ) pathways, frequently resulting in frameshift mutations that abolish JRAB protein expression.
This CRISPR knockout system enables efficient generation of MICALL2-deficient cell models for investigation of JRAB signaling, functional genomics studies, cancer biology research, and evaluation of therapeutic responses in human cell lines.
