JNK1 Antibody (37) is a mouse monoclonal IgG1 antibody raised against amino acids 264-415 of human JNK1, a stress-activated protein kinase critical for cellular stress responses. JNK1 mouse monoclonal antibody (37) recognizes an epitope within this specified region and demonstrates broad species cross-reactivity, detecting JNK1 in mouse, rat, human, and canine samples through Western blotting (WB), immunoprecipitation (IP), and immunofluorescence (IF) applications. The sophistication of JNK signaling becomes evident in its molecular architecture: the human JNK1 gene (PRKM8, SAPK1, MAPK8), positioned on chromosome 10q11.22, shares 83% amino acid identity with JNK2. Through complex alternative mRNA splicing mechanisms, three JNK genes generate ten distinct isoforms, including five p46 variants (JNK1α1, JNK1β1, JNK2α1, JNK2β1, JNK3α1) and five p54 variants (JNK1α2, JNK1β2, JNK2α2, JNK2β2, JNK3β2). JNK1′s dynamic subcellular distribution between cytoplasm and nucleus enables precise cellular responses to environmental stresses and inflammatory cytokines. Upon activation, JNK1 translocates to the nucleus, where JNK1 phosphorylates crucial transcription factors including c-Jun, ATF2, and Elk-1, thereby orchestrating stress-responsive gene expression patterns. This regulation proves particularly significant in immune function, where JNK1 facilitates the activation and differentiation of CD4+ helper T cells into TH1 and TH2 effector cells. JNK1 responds to diverse cellular stressors: capsaicin triggers JNK1 activation alongside p38 in Ras-transformed breast epithelial cells, while nitrogen oxides (NOx) induce coordinated upregulation of JNK1 with c-Fos, c-Jun, MEKK1, and p38. Through these sophisticated regulatory mechanisms controlling cell growth, differentiation, survival, and apoptosis, JNK1 (37) antibody serves as a valuable tool for investigating stress response pathways and cellular signaling mechanisms.

I am using JNK1 (37): sc-136205 for immunofluorescence, what dilution of this antibody should I use?

Asked by: Randy McDonald
Thank you for your question. We recommend a starting dilution of 1:50. However, optimal antibody concentration should be determined by titration. Please find our full immunofluorescence protocol here: https://www.scbt.com/scbt/resources/protocols/immunofluorescence-cell-staining
