Islet-2 Antibody (A-1) is a mouse monoclonal IgG1 kappa light chain antibody that detects Islet-2 protein of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). anti-Islet-2 antibody (A-1) is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. Islet-2 protein, also known as insulin gene enhancer protein ISL-2, consists of 359 amino acids and is encoded by the human gene ISL2. This nuclear protein features two N-terminal LIM domains, a homeodomain, and a serine/glutamine/threonine-rich C-terminus, which are crucial for function as a transcription factor. Islet-2 plays a vital role in defining subclasses of motor neurons in the spinal cord, influencing their segregation into distinct columns and guiding axon pathways. Islet-2 is initially expressed by all postmitotic spinal motor neurons, but expression is maintained in somatic motor neurons while being downregulated in visceral motor neurons during later embryonic stages. Early expression of Islet-2 in prospective visceral motor neurons is essential for the development of complete visceral motor neuron characteristics. Mutations that diminish or eliminate the activity of both Islet-1 and Islet-2 can lead to significant defects in the generation of visceral motor neurons and a loss of somatic motor neuron identity, underscoring the importance of Islet-2 in neuronal development and function.
