ISG15 Antibody (3E5) is a mouse monoclonal IgG1 antibody that detects ISG15 of human origin by western blotting (WB) and immunoprecipitation (IP). Anti-ISG15 antibody (3E5) is available as the non-conjugated form. ISG15 monoclonal antibody (3E5) was raised against a recombinant protein corresponding to amino acids 1-157 of ISG15 of human origin. The interferon-induced 15 kDa protein (ISG15) plays a crucial role in the immune response by functioning similarly to ubiquitin, conjugating to various intracellular target proteins, including JAK1 and MAPK3/ERK1, through a unique enzymatic pathway distinct from ubiquitin. This conjugation process regulates protein stability and activity, influencing numerous cellular processes such as inflammation and immune signaling. ISG15 (3E5) antibody exhibits specific chemotactic activity towards neutrophils, activating them to release eosinophil chemotactic factors, which further underscores ISG15′s importance in orchestrating immune responses. ISG15 (3E5) monoclonal antibody is predominantly expressed in muscle, epithelial, neuronal, and lymphoid cells, highlighting ISG15′s diverse roles in different tissue types. ISG15′s ability to induce interferon-gamma secretion from monocytes and macrophages also illustrates its involvement in paracrine, autocrine, and endocrine signaling mechanisms, making ISG15 a significant player in cell-to-cell communication within the immune system.

What blocking solution should I use with ISG15 (3E5): sc-69701 monoclonal antibody for Western blot?

Asked by: jenni
Thank you for your question. We recommend blocking with Blotto (either TBS Blotto A: sc-2333 or TBS Blotto B: sc-2335) Our Western blot protocol can be found here: https://www.scbt.com/scbt/resources/protocols/western-immuno-blotting
