IQGAP1 Antibody (F-7) is a mouse monoclonal IgG2a kappa light chain antibody that targets amino acids 314-422 near the N-terminus of the human IQGAP1 protein, allowing specific detection across human, mouse, and rat samples in applications like western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry with paraffin-embedded sections (IHCP), and enzyme-linked immunosorbent assay (ELISA). IQGAP1, or IQ motif containing GTPase activating protein 1, is a large, multi-domain scaffold protein with structural features that facilitate its role in signal transduction and cytoskeletal organization. Notably, IQGAP1 contains an N-terminal coiled-coil region thought to be involved in dimerization, a series of IQ motifs for calmodulin binding, and a C-terminal Ras GAP-related domain, although lacking GTPase-activating activity. This unique structure enables IQGAP1 to interact with multiple partners, including small GTPases Cdc42 and Rac1, linking IQGAP1 to actin regulation and cell morphology control. IQGAP1 also features a WW domain, enhancing binding to proline-rich sequences in target proteins, and regulates cell-cell adhesion through interactions with E-cadherin, β-catenin, and α-catenin at cell junctions. Calmodulin binding to IQGAP1, occurring in a calcium-dependent manner, disrupts association with Cdc42, thus fine-tuning IQGAP1′s activity in response to cellular signals.
