IL-10Rα Antibody (E-6) is a mouse monoclonal IgG1 kappa light chain antibody that detects IL-10Rα protein of mouse origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry with paraffin-embedded sections (IHCP), and enzyme-linked immunosorbent assay (ELISA). Anti-IL-10Rα antibody (E-6) is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. IL-10R is a crucial component of the immune system, belonging to the class II subgroup of the cytokine receptor family, and shares structural similarities with the interferon receptor. IL-10R is predominantly expressed in B cells and T helper cells, as well as in lipopolysaccharide-induced mouse fibroblasts, highlighting a role in immune response regulation. IL-10Rα is essential for mediating anti-inflammatory effects of interleukin-10 (IL-10), a cytokine that plays a significant role in limiting immune responses and preventing excessive inflammation. Upon IL-10 binding, IL-10R undergoes a conformational change that activates associated Janus kinases, JAK1 and Tyk2, leading to phosphorylation of specific tyrosine residues in the cytoplasmic domain of IL-10Rα. This post-translational modification is critical for IL-10R functionality and subsequent activation of IL-10 signaling pathways, which are vital for maintaining immune homeostasis. IL-10Rβ, which is ubiquitously expressed, forms heterodimeric receptors with both IL-22R and IL-28R subunits, expanding the functional repertoire of IL-10 signaling. IL-10R is also constitutively expressed on human natural killer (NK) cells, where direct binding of IL-10 enhances cytokine production, underscoring IL-10R importance in modulating immune responses.

Is this antibody recommended for detection of IL-10Rα of rat origin by IF?

Thank you for your question. IL-10Rα Antibody (E-6): sc-28371 has not yet been tested for reactivity against with IL-10Rα of rat origin. The antibody is raised against a recombinant protein immunogen that shares ~82% sequence identity with the published protein sequence for rat. There is a good chance that the antibody would react with rat. This antibody is recommended for IF, or immunofluorescent staining of fresh or frozen tissues or cells fixed in acetone or methanol. It has not yet been tested on formalin-fixed paraffin-embedded tissue sections. For a limited time, we are offering a free 10 µg sample of this antibody.
