IgM Antibody (G-12) is a mouse monoclonal IgG1 antibody that detects Immunoglobulin M (IgM) in human samples through various applications including western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry with paraffin-embedded sections (IHCP), and enzyme-linked immunosorbent assay (ELISA). IgM is the largest circulating antibody molecule in humans, consisting of a heavy chain (µ-chain) and a light chain (κ- or λ-chain), forming a pentameric structure that provides remarkable capacity for binding antigens due to ten binding sites, although steric hindrance limits simultaneous binding. IgM plays a crucial role in immune response, particularly during early stages of infection, as IgM is the first immunoglobulin expressed by mature B cells. IgM′s polymeric nature enhances avidity and facilitates effective complement activation, making IgM vital for opsonization and clearance of pathogens. Additionally, J chain presence allows IgM to function as a secretory immunoglobulin, contributing to mucosal immunity. While primarily found in serum, IgM′s large size restricts diffusion, resulting in low interstitial levels, which underscores IgM′s importance in maintaining immune homeostasis.
