IGF-IRβ Antibody (F-1) is a mouse monoclonal IgG2a kappa light chain antibody raised against amino acids 741-800 of human IGF-IRβ, also known as insulin-like growth factor 1 receptor beta or CD221. Anti-IGF-IRβ antibody (F-1) is validated for applications such as western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA), with reactivity in mouse, rat, and human species. IGF-IR is a receptor tyrosine kinase encoded by the IGF-IR gene on chromosome 15q26.3, and IGF-IR structure is essential for function in cellular signaling. IGF-IR receptor is composed of α and β subunits; α subunits are extracellular and responsible for ligand binding, while β subunits span the membrane and contain an intracellular tyrosine kinase domain. This structural organization allows IGF-IR to transmit signals from the extracellular environment to intracellular pathways, regulating key processes such as cell proliferation, differentiation, survival, and metabolism. IGF-IR′s modular structure enables interactions with proteins like Shc and insulin receptor substrate (IRS)-1 and -2, initiating signaling cascades such as the Ras-ERK and PI3K-AKT pathways. This functionality is critical for growth and development, and dysregulation of IGF-IR has been implicated in various cancers. Anti-IGF-IR antibody (F-1) serves as a valuable tool for exploring IGF-IR′s structural and functional roles in cell signaling and disease mechanisms.

Can IGF-IRβ (F-1): sc-390130 monoclonal antibody be used in IF or IHC with mouse tissue or cells? Will there be non-specific staining?

Thank you for your inquiry. The use of mouse monoclonal antibodies with mouse samples is very common and typically poses no problem. To eliminate any potential cross-reactivity of an anti-mouse secondary detection reagent, IGF-IRβ (F-1): sc-390130 is available in a variety of direct conjugations, such as HRP, PE, FITC and AlexaFluors.

Rated 5 out of 5 by from Highly potent and very specific antibodyThis monoclonal antibody was independently tested in-house by Kinexus Bioinformatics Corporation and validated for use for Western blotting. There was strong detection of the ~97 (beta-chain) and 200 kDa precursor forms of the target protein in an A431/HepG2/MCF7 cell lysate mix, but the target was not detected in mouse tissues (brain/testes/thymus/muscle mix). No major cross-reactivities were observed at 2 µg/ml antibody concentration with off-target proteins in the mouse tissues tested or in the A431/HepG2/MCF7 cell lysate mix.
