IFN-γRα Antibody (2E2) is a hamster monoclonal IgG antibody that detects IFN-γRα, also called interferon gamma receptor 1 or IFNGR1, in mouse, rat, and human samples through applications such as western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and flow cytometry (FCM). IFN-γRα Antibody (2E2) is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. The IFN-γ receptor, which includes IFN-γRα, plays a crucial role in mediating the effects of interferon-gamma (IFN-γ), a key cytokine involved in immune responses. Upon binding of IFN-γ, the receptor activates Janus kinases JAK1 and JAK2, leading to the phosphorylation of transcription factors Stat1α and Stat1β, which then translocate to the nucleus to initiate the transcription of genes involved in antiviral and immunomodulatory responses. Notably, while IFN-γRα is essential for ligand binding, IFN-γRα requires the presence of IFN-γRβ to fully activate the signaling cascade necessary for the expression of IFN-γ-inducible genes. The IFN-γRβ chain, also known as AF-1, is critical for signal transduction and varies in length between species, being 332 amino acids in mouse and 337 amino acids in human. This intricate interplay between the receptor components underscores the importance of both chains in orchestrating a robust immune response, making IFN-γRα a vital target for research in immunology and therapeutic interventions.

Thank you for your question. Yes, this antibody is applicable for Western blot (WB). If you purchase the HRP conjugated primary antibody (sc-12753 HRP), then no secondary antibody is needed for ECL detection.

Can IFN-γRα (2E2): sc-12753 monoclonal antibody be used in IF or IHC with mouse tissue or cells? Will there be non-specific staining?

Thank you for your inquiry. The use of mouse monoclonal antibodies with mouse samples is very common and typically poses no problem. To eliminate any potential cross-reactivity of an anti-mouse secondary detection reagent, IFN-γRα (2E2): sc-12753 is available in a variety of direct conjugations, such as HRP, PE, FITC and AlexaFluors.
