Mouse Ifi202b encodes IFI-202, an interferon-inducible member of the p200 (PYHIN) family implicated in innate immune regulation and control of cell fate decisions. IFI-202 has been linked to modulation of DNA damage responses, cell-cycle progression, and apoptotic signaling, with functional connections to interferon/ISG networks that shape inflammatory gene expression. In immune cells and interferon-stimulated contexts, altered Ifi202b activity is associated with dysregulated immune homeostasis and has been studied in relation to autoimmune-like phenotypes and inflammatory disease mechanisms in mouse models.
IFI-202 CRISPR/Cas9 KO Plasmid (m) is a pool of plasmids designed for targeted disruption of the Ifi202b gene in mouse cell lines. Each plasmid co-expresses a unique single guide RNA (sgRNA) targeting a distinct site within the Ifi202b together with the Streptococcus pyogenes Cas9 nuclease. The plasmids also encode GFP, allowing fluorescent identification and enrichment of successfully transfected cells by fluorescence microscopy or flow cytometry.
The multi-guide design increases the likelihood of generating insertions or deletions (indels) that disrupt the Ifi202b open reading frame following Cas9-mediated double-strand break formation. DNA breaks introduced by the CRISPR/Cas9 system are repaired through endogenous non-homologous end joining (NHEJ) pathways, frequently resulting in frameshift mutations that abolish IFI-202 protein expression.
This CRISPR knockout system enables efficient generation of Ifi202b-deficient cell models for investigation of IFI-202 signaling, functional genomics studies, cancer biology research, and evaluation of therapeutic responses in human cell lines.
