IDO Antibody (mIDO-48) is a rat monoclonal IgG2b antibody that detects Indoleamine 2,3-dioxygenase (IDO) in mouse, rat, and human samples through various applications including western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry, and enzyme-linked immunosorbent assay (ELISA). Anti-IDO antibody (mIDO-48) is available in both non-conjugated and multiple conjugated forms, such as agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and various Alexa Fluor® conjugates. Indoleamine 2,3-dioxygenase plays a crucial role in immune regulation by catalyzing the degradation of the essential amino acid L-tryptophan into N-formylkynurenine, a process that is vital for modulating T cell responses and maintaining immune tolerance. IDO′s activity is particularly important in the context of pregnancy, as IDO helps protect the developing fetus from maternal immune rejection by creating a local immunosuppressive environment. Additionally, IDO′s expression can inhibit T cell proliferation, which has implications for cancer immunotherapy and autoimmune diseases, making IDO a significant target for therapeutic intervention. The gene encoding human IDO is located on chromosome 8p12-p11, and IDO′s modulation through pharmacological or genetic means may provide strategies for controlling undesirable T cell responses in various clinical settings.

吉田と申します、質問失礼いたします。 マウスの細胞から抽出したタンパクをウエスタンブロッティングで解析するときにこちらの抗体を使用しております。この結果、50kと42kに日本のバンドが得られ、見本のようになりませんでした。この原因について関連がありそうな情報がありましたら教えていただけませんでしょうか。また、こちらの見本の抗体の希釈倍率など実験条件が知りたいです。 どうぞよろしくお願いいたします。

How does this Ab work for Human cell lines?

I noticed that sc-53978 is suitable for IHC, and i wondered that whether acidic or alkaline antigen retrieval solution is appropriate? Or maybe a protocol for IHC of sc-53978?

What is exact dilution

Thank you for your question! This antibody is supplied at 200 µg in 1 ml buffer. Dilution recommendations for different assays can be found in the datasheet available on the product page. The actual optimum dilution for your particular experimental setup and sample will have to be determined experimentally.

Whats the difference between this antibody and the IDO (H-11) sc-137012? The difference in the isotypes between then (IgG2b for the former and IgG1 for the latter) changes something in the secondary antibody that should be used for incubation?

sc-53978, used to do WB,Dilution ratio please?

Hi, we found three bands (close to 45 KDa) on our western blot after using this antibody (sample from rat brain). I wonder if it is different isoforms of IDO or something else. Was the full lenght protein it was raised against IDO1 or IDO2?

Thank you for your question. We have not tested this rat monoclonal antibody in rat brain samples. However, we would recommend that you include a secondary antibody control on your Western blots (omit primary antibody) to identify any possible non-specific binding from your anti-rat IgG secondary antibody alone in your rat brain samples. The exact epitope recognized by this rat monoclonal antibody has not yet been determined. It was raised against IDO1, and not IDO2 (also designated INDOL1).

Can you recommend a positive control to use with IDO Antibody (mIDO-48): sc-53978 for Western blot?

We recommend MOLT-4: sc-2233, Raji: sc-364236 or or human lymph node tissue extract: sc-363768 whole cell lysates to use as positive controls as we have Western blot data showing specific detection of IDO in these cell lines.

Rated 1 out of 5 by from QuestionableTried this antibody multiple times on mouse whole brain, hippocampus, striatum, lung and spleen. A 45 kDa band is present in all tissues, including tissues from Ido1 knock out mice. We have even found a similar band in cells that do not express Ido1 mRNA by qPCR. Has anyone confirmed this reagents specificity using Ido1 deficient samples, or am I the only one with this problem.
