SEE ALSO...

HPV16 L1 Antibody (CAMVIR-1) is a mouse monoclonal IgG2a kappa light chain antibody that detects HPV16 L1 protein of HPV-16 origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and immunohistochemistry with paraffin-embedded sections (IHCP). Anti-HPV16 L1 antibody (CAMVIR-1) is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. HPV16 L1 protein plays a crucial role in the viral life cycle, forming the major structural component of the virus′s capsid, which is essential for protecting and delivering viral genome into host cells. Pentameric assembly of L1 proteins creates a stable shell that facilitates initial attachment to host cell surface, a critical step in the infection process. C-terminus of L1 participates in both DNA binding and packaging, underscoring importance in viral replication cycle. Expression of L1 protein remains restricted to upper layers of infected epithelium, significant for understanding pathogenesis of HPV-related diseases, including cervical cancer. HPV16 L1′s ability to package unrelated plasmid DNA in vitro and deliver to eukaryotic cells highlights potential for gene therapy applications. Distinct localization patterns of L1 and interaction with minor capsid protein L2, found in punctate nuclear regions known as promonocytic leukemia protein oncogenic domains (PODs), provide insights into complex dynamics of HPV infection and cellular mechanisms involved.

I am using the LiCor/Odyssey system. Which secondary antibody do you recommend using with HPV16 L1 Antibody (CAMVIR-1): sc-47699

Asked by: jenni
We recommend using one of our our exclusive Mouse IgG Binding Proteins (m-IgGκ) conjugated to CFL 680 or CFL 790. They are suitable for use with Near-Infrared (NIR) detection systems, such as LI-COR/Odyssey and other comparable systems. A complete list of available binding proteins is available on our website here: https://www.scbt.com/scbt/browse/support-products-mouse-igg-binding-proteins/_/N-ecrety

Rated 1 out of 5 by from Antibody with issues! The Western Blot shown in the data sheet of “HPV16 L1 (CAMVIR-1): sc-47699, shows unspecific bands only, which are known to be seen with this antibody (Shafti-Keramat et al., J Virol. 2003 Dec;77(24):13125-35. PubMed PMID:14645569). There is no L1 signal depicted in the image (the correct size of L1 is markedly higher). As reported by Shafti-Keramat et al. it is an HPV unrelated dominant cellular band of app. 43 kDa. Furthermore it is known that HeLa cells are HPV type 18 positive (Lane A in the WB of the data sheet). We used this antibody, which gave us strong signals in the immunohistochemistry, implying the presence of HPV L1 in the tissue. This, however, could not be confirmed since the signal was derived from the unspecific HPV unrelated protein as seen in the Western blots. I therefore have to caution against working with this antibody, particularly when doing immuno(cyto)histochemistry. Using it in WB could potentially work since you at least could distinguish L1 from the unspecific band due to their different MW.
