HNF-3β Antibody (A-12) is a mouse monoclonal IgG1 kappa light chain antibody specific for an epitope mapping between amino acids 431-459 at the C-terminus of HNF-3β of mouse origin, exhibiting reactivity with mouse, rat, and human proteins by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA), and also reactive with additional species including equine, canine, bovine, and porcine. HNF-3β, also known as FOXA2 (Forkhead Box A2), is a critical transcription factor with a conserved forkhead DNA-binding domain that regulates genes essential for liver development, metabolism, and differentiation of endoderm-derived tissues. The function of HNF-3β is modulated by post-translational modifications such as phosphorylation, acetylation, and sumoylation, which influence transcriptional activity, stability, nuclear localization, and DNA-binding affinity. For instance, phosphorylation can alter HNF-3β′s interaction with other proteins and DNA, affecting gene expression patterns during development and in response to cellular signaling pathways. The hepatocyte nuclear factor family, comprising HNF-1 (α and β), HNF-3 (α, β, and γ), HNF-4 (α and γ), and HNF-6, regulates transcription of genes in the liver and other tissues like the kidney, small intestine, and thymus, with HNF-3α, HNF-3β, and HNF-3γ involved in controlling numerous hepatocyte genes in the adult liver and playing roles in gastrulation events such as body axis formation. HNF-6 binds to the promoter of HNF-3β, indicating a potential role in gut endoderm epithelial cell differentiation, and may act as a transcriptional activator for multiple hepatocyte-enriched genes, including cytochrome P450 2C13 and α-1 antitrypsin, making anti-HNF-3β antibody (A-12) a valuable tool for researchers studying the role of HNF-3β and post-translational modifications in various biological processes and diseases such as diabetes, liver dysfunction, and cancer.
