HMGCR Antibody (C-1) is a mouse monoclonal IgG1 kappa light chain antibody that detects HMGCR protein of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry, and enzyme-linked immunosorbent assay (ELISA). Anti-HMGCR antibody (C-1) is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. The human enzyme hydroxy-3-methylglutaryl coenzyme A reductase (HMGCR) plays a critical role in regulating cholesterol synthesis, which is essential for cellular growth and membrane integrity, particularly in liver tissue. HMGCR activity is tightly controlled through phosphorylation, a post-translational modification that inactivates HMGCR in response to sterols and non-sterol metabolites, thereby preventing excessive cholesterol production. This regulatory mechanism is vital for maintaining lipid homeostasis and preventing conditions such as hypercholesterolemia. HMGCR serves as the target of statins, which are widely used to lower plasma cholesterol levels and reduce the risk of cardiovascular events, highlighting HMGCR′s importance in both metabolic regulation and therapeutic interventions. Intermediates produced in the cholesterol synthesis pathway can influence signaling pathways, such as those involving Ras, suggesting that HMGCR regulation may have implications for novel chemotherapeutic strategies.

Does this antibody recognize HMGCR produced by zebrafish? Are there any studies confirming this interaction?

Thank you for your question. We are not aware of any publications citing use of this antibody, HMGCR (C-1): sc-271595, in zebrafish. The antibody is raised against amino acids 589-888 mapping at the C-terminus of HMGCR of human origin. If you can provide the accession number or protein sequence for the zebrafish protein we could perform sequence analysis to try and predict if the antibody may cross-react with this species. When would it be better to use sc-271595 AC instead of sc-271595?

Thank you for your question. If the only application you intend to use this HMGCR antibody for is IP, then the agarose conjugated form sc-271595 AC is best. If multiple applications are required, then the unconjugated form sc-271595 is best.

Application: Western blot Sample: whole lysate of human hepatocytes from Huh7 cell line Gel running conditions: reduced, denaturing, 4-12% SDS-PAGE Loading amount: 30 µg protein/lane Transfer: semi-dry, on nitrocellulose membrane (0.45 µm) Blocking agent: 5% milk in TBST buffer, for 1h, RT Primary antibody dilution- 1: 500 in 5% milk, for 16 h at 4°C Secondary antibody: non- Santa Cruz antibody anti-mouse-HRP, 1:10000 dilution in 5% milk, for 1h, RT Detection: ECL substrate (femto sensitivity) Exposure time: 1 sec Bands: a strong, band at approx. 60 kDa (probably the HMGCR C-terminal cleavage products); fade, non-specific bands at 35 kDa.

Rated 1 out of 5 by from Did not work in WBI used this Ab in WB using lysates from CHO cells (chinese hamster ovary). Although the product is recommended, there was no band detected in the range of 90-130 kD. There were only fuzzy, non-specific bands at 230 kD. Rest of the antibodies I used in the same blot worked just fine. I did not use it for IF.
