SEE ALSO...

HLA-DOβ Antibody (DOB.L1) is a mouse monoclonal IgG2b kappa light chain antibody that detects the HLA-DO beta protein of human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and immunohistochemistry with paraffin-embedded sections (IHCP). HLA-DOβ (DOB.L1) antibody is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. HLA-DO beta protein plays a crucial role in the immune response by regulating the presentation of antigens to CD4+ T cells, which is essential for initiating adaptive immunity. Specifically, HLA-DO beta protein interacts with HLA-DM to facilitate the exchange of peptides in the antigen-binding groove of major histocompatibility complex (MHC) class II molecules. This interaction is vital for ensuring that T cells can recognize and respond to foreign antigens effectively. Proper functioning of HLA-DO beta protein is particularly important in the context of autoimmune diseases, as variations in the genes encoding these molecules have been linked to susceptibility to conditions such as rheumatoid arthritis. Using anti-HLA-DOβ antibody (DOB.L1) in research can provide insights into the mechanisms of antigen presentation and the role of HLA molecules in immune regulation.

Does this antibody bind the native or denatured HLA-DOB protein?

Asked by: Dortac
Thank you for your question. If the denatured form contains the cytoplasmic portion of the protein sequence than it should detect both. For further assistance please call 800-457-3801.

I am using HLA-DOβ (DOB.L1): sc-69739 monoclonal antibody to stain paraffin embedded tissue. Do you recommend antigen retrieval when using this antibody?

Asked by: Professor Griffin
We recommend performing antigen retrieval with sodium citrate buffer (pH 6) and heat. The full protocol can be found here: https://www.scbt.com/scbt/resources/protocols/immunoperoxidase-staining
