HIV-1 Nef Antibody (JR6) is a mouse monoclonal IgG1 antibody that detects Nef protein of HIV-1 origin by western blotting (WB), immunofluorescence (IF), and immunohistochemistry with paraffin-embedded sections (IHCP). HIV-1 Nef (JR6) antibody is available in a non-conjugated form, providing researchers with a versatile tool for studying Nef protein′s critical role in HIV pathogenesis. Nef, or negative factor, is a peripheral membrane protein that plays a pivotal role in manipulating host cell signaling pathways and evading immune response. Nef′s interaction with Src-family tyrosine kinases and GTPase activators occurs at its SH3-binding domain, which is crucial for modulating phosphoinositide 3-kinase sphingomyelinase signaling pathways, ultimately leading to CD4 downregulation on T cells. By promoting rapid endocytosis of CD4 from the cell surface, Nef aids in virus evasion of the immune system and disrupts normal immune response, making this protein a significant factor in HIV infection progression. Nef′s ability to alter transmembrane protein distribution and inhibit MHC class II-restricted peptide presentation reduces surface expression of mature MHC class II while increasing levels of immature MHC class II associated with invariant chains. As the only HIV-1 gene product with such an effect, Nef plays a critical role in viral lifecycle and host immune evasion strategies.
