HIV-1 Nef Antibody (2F2) is a mouse monoclonal IgG1 antibody that detects HIV-1 Nef in HIV-1 by applications such as western blotting (WB), immunofluorescence (IF), and immunohistochemistry. HIV-1 Nef is a crucial peripheral membrane protein that plays a significant role in the pathogenesis of HIV by modulating host cell signaling pathways and immune responses. Anti-HIV-1 Nef antibody (2F2) recognizes a protein that interacts with various cellular proteins, particularly Src-family tyrosine kinases and GTPase activators, through its SH3-binding domain. This interaction is vital for HIV-1 Nef′s function in manipulating the host′s immune system, as HIV-1 Nef facilitates the downregulation of CD4, a critical receptor on helper T cells, by promoting its rapid endocytosis. HIV-1 Nef′s structure includes two relatively unstructured loops that are essential for engagement with proteins involved in membrane trafficking, allowing control over the distribution of transmembrane proteins and evasion of immune detection. By inhibiting MHC class II-restricted peptide presentation, HIV-1 Nef effectively reduces the surface expression of mature MHC class II molecules while increasing levels of immature MHC class II associated with invariant chains. This unique capability makes HIV-1 Nef the only HIV-1 gene product that can perform such immune evasion, underscoring its importance in the viral life cycle and the challenges for therapeutic interventions.
