HES5 Antibody (3B6) is a mouse monoclonal IgG2a kappa light chain antibody that detects HES5 protein of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), and enzyme-linked immunosorbent assay (ELISA). Anti-HES5 antibody (3B6) is available as the non-conjugated format. HES5 is a member of the basic helix-loop-helix (bHLH) transcriptional repressor family, which plays a crucial role in the Notch signaling pathway, essential for proper segmentation and neural development during embryogenesis. HES5 is predominantly located in the nervous system, where HES5 regulates the expression of genes involved in neurogenesis and myogenesis. HES5 interacts with TLE proteins, the mammalian homologues of Drosophila Groucho, through its carboxy-terminal WRPW motif, forming a complex that binds directly to DNA, thereby repressing transcription without interfering with activator proteins. HES5 preferentially binds to the N box (CACNAG) DNA sequence, which is critical for its function in gene regulation. HES5 expression is tightly regulated during development, and dysregulation can lead to various neurodevelopmental disorders, highlighting HES5′s importance in maintaining proper cellular functions and developmental processes.
