HDAC9 Antibody (B-1) is a mouse monoclonal IgM antibody that detects HDAC9 protein of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). Anti-HDAC9 antibody (B-1) is available as the non-conjugated form. HDAC9 protein plays a crucial role in gene expression regulation through function as a histone deacetylase, which removes acetyl groups from lysine residues on histones, leading to chromatin condensation and transcriptional repression. This process is vital for maintaining cellular homeostasis and regulating various biological processes, including cell differentiation and proliferation. HDAC9 is involved in muscle gene expression modulation and has been implicated in various diseases, including cancer and neurodegenerative disorders. HDAC9 exists in two alternatively spliced isoforms, HDAC9 and HDAC9a, with both retaining HDAC catalytic domain and deacetylase activity, yet differing in expression patterns and functional roles. HDAC9 is predominantly expressed in tissues such as brain, skeletal muscle, kidney, placenta, and pancreas, highlighting significance in both normal physiology and disease states.

Has this antibody been validated recently by blotting HDAC9 KO cell lysate, or ectopic HDAC9 protein?

Thank you for your question. We have not tested this mouse monoclonal antibody, HDAC9 (B-1): sc-398003, using an HDAC9 knockout cell line. The antibody has been validated using a variety of tests, including the Western blot data published on our website in Ramos and NAMALWA whole cell lysates. We do offer a range of human specific HDAC9 CRISPR products that could be used in combination with this antibody as controls to demonstrate antibody specificity to the target protein. Rated 1 out of 5 by from only detected 55kda bandUsing human blood monocyte derived macrophages, 25 ug total cell lysate ran on midi gels, 1:500 dilution in milk, 1:4000 2nd Ab CST anti-mouse we could only detect a band at wrong size. The signal was also not too strong.

Rated 1 out of 5 by from this HDAC9 antibody is kind of not working wellI got a free sample of the HDAC9 antibody from Santa Cruz and tested it for Western blotting. A clean and strong band with a size around 140 kD did appear. But the band is kind of not for HDAC9. Because the antibody even cannot recognize overexpressed Flag-tagged full HDAC9, which, however, can be detected by anti-Flag or some other anti-HDAC9 antibodies. Does any other person get a similar result? Can you help to recommend a good HDAC9 antibody to detect endogenous HDAC9 by WB and IP, etc?
