    SEE ALSO...

HCR Antibody (E-4) is a mouse monoclonal IgG1 kappa light chain antibody that detects HCR protein of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry, and enzyme-linked immunosorbent assay (ELISA). Anti-HCR antibody (E-4) is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. HCR protein, also known as coiled-coil α-helical rod protein 1 or putative gene 8 protein, consists of 782 amino acids and is encoded by the human gene CCHCR1. HCR protein plays a crucial role in regulating keratinocyte proliferation and differentiation, which is vital for maintaining skin integrity and function. HCR protein is predominantly located in the nucleus and is highly expressed in tissues such as the heart, liver, skeletal muscle, kidney, and pancreas, with lower expression levels in the lung and placenta. Notably, HCR protein is overexpressed in keratinocytes of psoriatic lesions, linking HCR to psoriasis, a chronic inflammatory skin condition affecting around 2% of the population. Psoriasis is characterized by red, scaly skin lesions, often found on the scalp, elbows, and knees, and can be associated with severe arthritis. The hyperproliferation of keratinocytes and the infiltration of inflammatory cells into the dermis and epidermis contribute to the formation of these lesions. While the usual onset of psoriasis occurs between the ages of 15 and 30, psoriasis can manifest at any age. The association of HCR with psoriasis appears to be due to linkage disequilibrium with the CW 0602 allele, although HCR is unlikely to be directly involved in the disease′s development.

Can HCR (E-4): sc-365889 monoclonal antibody be used to stain formalin-fixed, paraffin-embedded (FFPE) tissue sections?

    Asked by: jerojero
Thank you for your question. Yes, HCR (E-4): sc-365889 has been validated for use in IHC with paraffin-embedded sections. We recommend performing antigen retrieval with sodium citrate buffer (pH 6) and heat. The full protocol can be found here: https://www.scbt.com/scbt/resources/protocols/immunoperoxidase-staining
