HCP1 Antibody (B-4) is a mouse monoclonal IgG1 kappa light chain antibody that detects HCP1 protein of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry, and enzyme-linked immunosorbent assay (ELISA). HCP1 Antibody (B-4) is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. HCP1, also known as heme carrier protein 1 or proton-coupled folate transporter (PCFT), is a multi-pass transmembrane protein predominantly expressed in the small intestine, particularly localized to the apical brush border of the duodenum and jejunum. HCP1 plays a crucial role in folate absorption, which is essential for DNA synthesis and repair, making HCP1 vital for cellular function and overall health. HCP1 operates optimally at a low extracellular pH of approximately 5.5, facilitating folate transport independently of sodium ions and membrane potential. HCP1 exhibits a high affinity for folic acid while showing low affinity for PT523 antifolate. Notably, HCP1 undergoes post-translational regulation by iron levels in the duodenum; during iron deficiency, HCP1 localizes to the apical membrane, whereas excess iron causes HCP1 to shift to the cytoplasm. Additionally, sulfasalazine acts as a potent inhibitor of HCP1, and mutations in the HCP1 gene can lead to hereditary folate malabsorption, an autosomal recessive disorder.
